Analogues of growth hormone-releasing hormone, acting on a known receptor with a well-characterised downstream axis. Mechanistically the least speculative family in the library — the uncertainty here is about clinical benefit rather than about what the molecule does.
Four compounds, one receptor, and four different evidence positions. That gap is the reason this family is worth reading as a set.
Tesamorelin and Sermorelin went through approval pathways, which is what produces characterised human pharmacokinetics and a stated indication. CJC-1295 with DAC has two published human studies plus a pulsatility substudy. CJC-1295 No-DAC has no identified human pharmacokinetic data and no randomised trial, despite being sold and discussed alongside the other three. Acting on the same receptor as an approved drug is a mechanistic similarity, not a transfer of evidence.
