Hypothalamic and pituitary signalling peptides, plus one sleep peptide. Every compound here acts on a known receptor with established human pharmacology — the uncertainty is entirely about whether the downstream effects amount to a clinical benefit.
The newest human sleep evidence in this family is more than thirty years old.
DSIP‘s six located human sleep trials all fall between 1981 and 1992. The only modern human study found for the guide used it as an adjunct to isoflurane anaesthesia. Intranasal oxytocin carries Mixed Evidence with a retracted meta-analysis in its own record — 2015, retracted 2018 for data-entry and analytic errors — no improvement in any aspect of symptoms across eight randomised trials in schizophrenia, and a finding that repeated administration does not improve most core symptoms in autism. Gonadorelin is the family’s approved compound, and its central pharmacological fact runs in both directions: pulsatile delivery stimulates the axis, sustained agonism suppresses it.
