Lyophilisation, cold chain and what degrades in transit

01 · Why peptides ship freeze-dried

Peptides in solution degrade. Water enables hydrolysis of the peptide bond, oxidation of susceptible residues, and aggregation. Removing the water slows all three by orders of magnitude, which is why research peptides ship as a lyophilised cake rather than a liquid.

Lyophilisation freezes the solution and removes water by sublimation under vacuum, so the material never passes back through a liquid phase. What remains is a porous solid that reconstitutes readily.

02 · What the cake tells you

A lyophilised cake has a normal appearance: uniform, occupying roughly the volume it was frozen in, intact rather than collapsed.

03 · What a temperature excursion actually does

Degradation kinetics are temperature-dependent, so the meaningful quantity is not whether material got warm but how warm, for how long. A short excursion during transfer is different from a package sitting on a loading dock over a weekend.

Lyophilised material is far more tolerant of this than solution, which is the point of shipping it dry. Tolerant is not immune, and the tolerance is not usually quantified for a given compound because the stability study establishing it has not been done.

04 · Which claims are checkable and which are not

Checkable after the fact: the stated storage condition on the label, whether a temperature logger was included and what it recorded, the state of the cake on arrival, and the packaging itself — insulation and coolant either present or not.

Not checkable after the fact: “cold chain maintained”, “shipped on ice”, and every unlogged assurance about what happened between two buildings. These are not necessarily false. They are simply not evidence, and should not be filed as though they were.

05 · Storage and reconstitution

A supplier stating storage conditions specifically — a temperature for the sealed vial, a different one after reconstitution, and an expected in-use period — is describing a product they have thought about. One stating “store cool” is describing a product they have not.

The same applies to reconstitution: which diluent, what volume, and how to handle the solution afterwards. Stated conditions are checkable. Implied ones are not.

None of this addresses whether a compound does anything. Handling determines whether the material is what the certificate said it was by the time it arrives — a narrower question, and the only one shipping can answer.
